Have 1992 Yamaha outboard VX 76degree TXRQ model 250 hp, been a few years since I replaced the impeller, watched a video on youtube but wasn't clear on weather outboard should be in gear when lower unit is removed, any
    suggestions

I find it easier for it to be in neutral, but it doesnt matter, just make sure the shift control handle matches the actual position of the shift in the lower unit, also put 1 bolt in and check the shifter, it will save you lots of time and grief..

I would keep everything in neutral.

There's NO reason to shift the control lever nor to mess with the shifter in the LU. It'll come apart with the shifter lined up, it'll go back together all lined up (if not messed with)..
